After a very challenging summer, I've finally turned in my written thesis, so it's official: I have my Ph.D. I'm publishing the abstract below. These findings should all be published in peer-reviewed journals in the next 6 months.


Ataxin-7 Conserved Motifs Determine the Severity of the Neurodegenerative Disorder Spinocerebellar Ataxia Type 7 in Transgenic Mice and Influence Lifespan in Yeast

Spinocerebellar ataxia type 7 (SCA7) is an autosomal dominant, progressive neurodegenerative disorder whose characteristic features are cerebellar ataxia, dysarthria, and retinal cone-rod dystrophy culminating in blindness. SCA7 is caused by an abnormally long glutamine-coding CAG repeat in the SCA7 gene, which encodes the protein Ataxin-7.

Ataxin-7 contains several conserved motifs that may influence the toxicity of the glutamine tract. Among these are three conserved regions (conserved block I – III), two caspase-7 cleavage sites, a nuclear export signal and two monopartite nuclear localization signals (NLS). Previous investigations have shown that the caspase-7 cleavage site D266 is required for the full toxicity of the Ataxin-7 protein in cell culture. We generated SCA7 transgenic mice expressing a 92 CAG version of the human SCA7 cDNA, with and without a D266N mutation. Mice carrying the D266N mutation were protected from SCA7-like neurodegeneration, behavioral signs and shortened lifespan.

To further characterize the role of conserved motifs in SCA7 pathology, we generated SCA7 transgenic mice carrying point mutations in both C-terminal NLSs (KKRK -> KAAK). Previous work has shown that nuclear localization is an important step in the pathology of CAG repeat disorders. We observed that mice lacking C-terminal NLS activity were substantially protected from degeneration of the retina and cerebellum, SCA7-like behavioral signs and shortened lifespan.

Age is the primary risk factor for neurodegenerative disease. Even in the absence of overt disease, the aging brain shows histopathological and molecular changes reminiscent of neurodegeneration. To explore the link between neurodegenerative disease and aging, we have examined the replicative lifespan of Saccharomyces cerevisiae missing the SCA7 ortholog, SGF73. This strain exhibits an unusually long lifespan, which is dependent on the function of the NAD+-dependent deacetylase SIR2. We present evidence that the extended lifespan of the SGF73 null strain is due to the influence of Sgf73 on the activity of Sir2 and the histone deubiquitinase Ubp8. Furthermore, we show that the level of ubiquitinated H2B is elevated in an SCA7 transgenic mouse line, indicating that an alteration in Ubp8 activity may play a role in SCA7 pathology and that aging and neurodegeneration may share a common mechanism.

Today, Film Studies For Free is very merrily celebrating its first birthday. It is marking this auspicious date -- in style and on style -- by posting one of its longest links lists yet: to online and openly accessible articles and essays on the subject of film criticism (scholarly and otherwise) that FSFF's author has found important and/or stimulating over the last years.

FSFF would like its list to be even longer, though, so do please take note of the four headings below (on film style: on film criticism; on film critics; and important, self-reflexive, examples of film criticism online) and do let the blog know of links to other relevant work (especially to good examples of online film criticism), preferably in the comments section, please.

This post was inspired, in great part, some weeks back by the peerless Girish Shambu who launched a characteristically thoughtful and important discussion, in a blog entry entitled "Building A Large Conversation", about the divide that exists between the fields of film scholarship and film criticism. Girish wrote:

Except for a small number of invaluable critic-scholars who work to bridge the gap, the two groups similarly shy away from citing each other. Why is this so? For critics, it would require the significant effort of familiarizing themselves with scholarly literature past and present, an effort made more difficult by the presence of a specialized scholarly vocabulary. For scholars, whose jobs already require them to do vast amounts of reading, this would mean widening their field of vision to include writing in film magazines, the Internet (including blogs), and newspapers. Added to this are the demands in both professions of watching scores of films on a steady basis.


Like Girish, the many important commenters to his blog post, and other thoughtful respondents to it, such as HarryTuttle, FSFF readily acknowledges the difficulties in bridging these gaps.

In his response to Girish's post, film scholar and blogger Chris Cagle wrote eloquently and concisely about those difficulties, but in an optimistic frame, he noted that what might be needed is

a model that's different than pure specialization or pure dilettantism. For lack of a better name, I'd call it randomization. Each scholar specializes but looks to new ideas, methodologies, and inspiration in a limited fashion with the hope that collectively we mitigate the downside of stale intellectual mindsets. The journalist, blogger, or public intellectual could have a role in this.

Film Studies For Free owes its very existence to the desire to help to 'join up' scholars and critics in the global online arena. And it very much seconds Cagle's assertions about what is required to achieve this. Today, then, it reaffirms its own mission by helping to encourage a richer and more connected 'scholar-critic conversation' through the below list of 'randomly collected' but also 'specialized' links.

On a final note, FSFF has received lots of encouragement in its first year of existence, but none warmer, more timely or more generous than that given in its early days by Girish, whose own website continues to be a huge inspiration in all sorts of ways. Giving up a year of my life

To save the rest of it.

That's what the doctor told me I have to do to cure this cancer.

I have a daughter that will need me in her life, so that is the plan.

I spent the past week at Sloan. Taking tests. Speaking with doctors.

And crying... a lot of crying.

It is a surreal experience. Trying to understand the complicated schedules and make sense of how my family can manage life without mom.

And not just without mom. Life with a cancer patient.

I'll be treated as an outpatient at Sloan on a clinical trial.

My treatments with be 3 week cycles of chemotherapy for about a year. Typically the cycles are 2 weeks of treatment and 1 week of rest. During my week of rest, I will have to go in for tests.

When I have radiation, it will be 6 weeks with no break. This does not change the chemo schedule.

Some treatments are an hour. Some are ten.

I live about 1.5 hours away from Sloan with no traffic.

So every day is 3 hours longer, just for driving. Probably a little more.

3 hours of being stuck in the car and potentially very ill.

My initial reaction was defeatism.

My next reaction was complete and total fear.

I have a daughter and husband who need me. Who love me.

I am determined to make this work.

I will stay in a hotel on the long days.

I will coordinate a schedule of driving with my family and friends. I hope to have enough people to call when I cannot make it to the city and someone will help.

In the event that falls through, I will use a car service. The sustainability of this option is low for its cost and the emotional drain of being alone. I hope it is a rare thing.

I will sleep in my own bed. I will get to see my daughter and husband everyday.

I will remain positive and strong.

This year of my life. The year I had cancer. The year before I was a cancer survivor starts now.
